RESUMEN

Panicle type is one of the important factors affecting rice (Oryza sativa L.) yield, and the identification of regulatory genes in panicle development can provide significant insights into the molecular network involved. This study identified a large and dense panicle 1 (ldp1) mutant produced from the Wuyunjing 7 (WYJ7) genotype, which displayed significant relative increases in panicle length, number of primary and secondary branches, number of grains per panicle, grain width, and grain yield per plant. Scanning electron microscopy results showed that the shoot apical meristem (SAM) of ldp1 was relatively larger at the bract stage (BM), with a significantly increased number of primary (PBM) and secondary branch (SBM) meristematic centers, indicating that the ldp1 mutation affects early stages in SAM development Comparative RNA-Seq analysis of meristem tissues from WYJ7 and ldp1 at the BM, PBM, and SBM developmental stages indicated that the number of differentially expressed genes (DEGs) were highest (1407) during the BM stage. Weighted gene coexpression network analysis (WGCNA) revealed that genes in one module (turquoise) are associated with the ldp1 phenotype and highly expressed during the BM stage, suggesting their roles in the identity transition and branch differentiation stages of rice inflorescences. Hub genes involved in auxin synthesis and transport pathways, such as OsAUX1, OsAUX4, and OsSAUR25, were identified. Moreover, GO and KEGG analysis of the DEGs in the turquoise module and the 1407 DEGs in the BM stage revealed that a majority of genes involved in tryptophan metabolism and auxin signaling pathway were differentially expressed between WYJ and ldp1. The genetic analysis indicated that the ldp1 phenotype is controlled by a recessive monogene (LDP1), which was mapped to a region between 16.9 and 18.1 Mb on chromosome seven. This study suggests that the ldp1 mutation may affect the expression of key genes in auxin synthesis and signal transduction, enhance the size of SAM, and thus affect panicle development. This study provides insights into the molecular regulatory network underlying rice panicle morphogenesis and lays an important foundation for further understanding the function and molecular mechanism of LDP1 during panicle development.

Anthropogenic emissions have resulted in increases in the atmospheric fluxes of both nutrient and toxic elements. However, the long-term geochemical impacts on lake sediments of deposition activities have not been clearly clarified. We selected two small enclosed lakes in northern China-Gonghai, strongly influenced by anthropogenic activities, and Yueliang lake, relatively weakly influenced by anthropogenic activities-to reconstruct historical trends of atmospheric deposition on the geochemistry of the recent sediments. The results showed an abrupt rise in the nutrient levels in Gonghai and the enrichment of toxic metal elements from 1950 (the Anthropocene) onwards. While, at Yueliang lake, the rise on TN was from 1990 onwards. These consequences are attributable to the aggravation of anthropogenic atmospheric deposition in N, P and toxic metals, from fertilizer consumption, mining and coal combustion. The intensity of anthropogenic deposition is considerable, which leave a significant stratigraphic signal of the Anthropocene in lake sediments.

BACKGROUND: COVID-19 is still soaring, and the new delta COVID-19 variant is on the rise and spreading around the world. OBJECTIVE: We conducted a patent analysis to better understand the therapeutic strategy developed for antivirals available for the disorders of the respiratory system. MATERIALS AND METHODS: European granted patents filed from January 2002 to June 2021 were analyzed. We used a combination of International patent classification (IPC) "A61p31/12" and "A61p11/00" to search the relevant documents. RESULTS: Our study showed R&D of antiviral drugs for disorders of the respiratory system to be decreasing over the past 20 years. Chemical drugs showed various chemical structures. The development of chemical drugs or herbal medicines appeared to commence earlier than the biological products. Also, the results indicated that large global companies play a leading role in developing kinase inhibitors as chemical drugs. CONCLUSION: There are three strategies for developing antiviral drugs for the disorders of the respiratory system, including chemical drugs, herbal medicines or natural products, and biological products. Herbal medicines may provide a new insight and approach to developing antiviral drugs for disorders of the respiratory system. A combination of chemical drugs and natural products may be a promising therapeutic method for treating patients with COVID- 19.

Global climate change and human activities have significantly impacted lake ecosystems at an accelerating rate in recent decades, but the differences between the responses of lake ecosystems to these two stressors remain unclear. Thus an improved understanding of the long-term influences of climatic and anthropogenic disturbances is necessary for the management of lake ecosystems. In order to address these issues, a sedimentary record was obtained from Lake Yilong in Yunnan Province in southwestern China, where the climate and natural environment are dominated by the Indian Summer Monsoon and there is a long history of human occupation and intensive human activity. The chronology is based on AMS 14C dates from 13 samples of plant macrofossils and charcoal, which show that the record spans the last ~12,000 yr. Geochemical indices were used to reconstruct hydro-climatic variations and lake ecosystem responses. The results indicate that a cold and humid climate prevailed from the late Pleistocene to the beginning of the Holocene, which was interrupted by an abrupt decrease in precipitation during 9.7-8.7 ka (1 ka = 1000 cal yr BP, corresponding to the 9.3 ka event). A persistent drying trend occurred during the middle and late Holocene, and there was an increase in the intensity of human activity during the past 1500 years. A comparison of the effects of a natural climatic event and human disturbance reveals contrasting lake ecosystem responses. The lake ecosystem was resilient to the 9.3 ka event and subsequently recovered; however, long-term human activity in the watershed, including deforestation and cultivation, reduced the stability of the lake ecosystem and positive feedback effects were strengthened, leading to the deviation of the system far from its previous stable state. It is concluded that, compared to climate change, human activities have had a much more serious impact on lake ecosystem.

Gastric cancer is one of the major causes of cancer-associated mortality worldwide. miR-1307-3p has been demonstrated to serve multiple roles in the development of various types of cancer. The present study aimed to evaluate the expression and functional role of miR-1307-3p in the progression of gastric cancer. The expression of miR-1307-3p in gastric cancer tissues and cell lines was detected by reverse transcription quantitative PCR. Furthermore, the correlation between miR-1307-3p expression and the clinicopathological characteristics and prognosis of patients was evaluated. Cell Counting Kit-8 and Transwell assays were performed to analyze the effects of miR-1307-3p on the proliferation and the migratory and invasive abilities of gastric cancer cells, respectively. Dual-luciferase reporter assay was conducted to reveal the potential underlying mechanism of miR-1307-3p. In gastric cancer tissues and cells, miR-1307-3p expression was significantly upregulated compared with the normal tissues and cell lines. In addition, the expression of miR-1307-3p was associated with the Tumor-Node Metastasis stage of patients. The results from Cox regression analysis demonstrated that miR-1307-3p may serve as an independent predictor for the prognosis of patients with gastric cancer. Furthermore, the upregulation of miR-1307-3p in gastric cancer cell lines significantly promoted the cell proliferation and migratory and invasive abilities by targeting DAB2 interacting protein. In conclusion, the findings from the present study suggested that miR-1307-3p may serve as a tumor promoter of gastric cancer and that miR-1307-3p expression in tumor tissues may be used as a prognostic indicator for patients with gastric cancer.

Neuronal ceroid lipofuscinoses (NCLs) are a group of autosomal recessive inherited neurodegenerative disorders mainly affecting children, and at least 13 causative genes (CLN1 to CLN8 and CLN10 to CLN14) have been identified. Here, we reported a novel homozygous missense mutation (c.434G > C, p.Arg145Pro) identified in CLN5 gene via whole exome sequencing in a 5-year-old girl. The patient first presented paroxysmal epilepsy associated with vomiting, followed by progressive regression in walking, vision, intelligence and speaking. Combining the molecular and clinical analysis, the diagnosis of NCL could be made, although the missense mutation (c.434G > C, p.Arg145Pro) in CLN5 was evaluated to be a variant of uncertain significance according to American College of Medical Genetics and Genomics (ACMG) standard. We further performed expression and localization studies and our results provide evidence of impaired cellular trafficking of CLN5 to lysosome, indicating that this mutation might be deleterious to the function of CLN5 for its mislocalization. Our study demonstrated the efficacy of next generation sequencing in molecular diagnosis, and a deleterious effect of the variant discovered in our patient on CLN5, triggering the NCL disease.

Changes in global climate intensify the hydrological cycle, directly influence precipitation, evaporation, runoff, and cause the re-distribution of water resources in time and space. The aridity index (AI), defined as the ratio of annual precipitation to annual potential evapotranspiration, is a widely used numerical indicator to quantify the degree of dryness at a given location. This study examined the effects of climate change on AI in China during 1961-2015. The results showed that the nationally averaged AI experienced a notable interdecadal transition in 1993, characterized by increasing AI (wetter) between 1961 and 1993, and decreasing AI (drier) after 1993. Overall, the decreased solar radiation (solar dimming) was the main factor affected the nationally averaged AI during 1961-1993, while the relative humidity dominated the variations of nationally averaged AI during 1993-2015. However, the roles of individual factors on the changes in AI vary in different subregions. Precipitation is one of the important contributing factors for the changes of AI in almost all subregions, except the Mid-Lower Yangtze and Huaihe basins. Solar radiation has been significantly decreased during 1961-1993 in South China, Southwest China, Mid-Lower Yangtze and Huaihe basins, and the Tibetan Plateau. Therefore, it dominated the trends of AI in these subregions. The relative humidity mainly affected the Mid-Lower Yangtze and Huaihe basins, Southwest China, and the Tibetan Plateau during 1993-2015, hence dominated the trends of AI in these subregions. The changes of temperature and wind speed, however, played a relatively weak role in the variations of AI.

The lack of a precisely-dated, unequivocal climate proxy from northern China, where precipitation variability is traditionally considered as an East Asian summer monsoon (EASM) indicator, impedes our understanding of the behaviour and dynamics of the EASM. Here we present a well-dated, pollen-based, ~20-yr-resolution quantitative precipitation reconstruction (derived using a transfer function) from an alpine lake in North China, which provides for the first time a direct record of EASM evolution since 14.7 ka (ka = thousands of years before present, where the "present" is defined as the year AD 1950). Our record reveals a gradually intensifying monsoon from 14.7-7.0 ka, a maximum monsoon (30% higher precipitation than present) from ~7.8-5.3 ka, and a rapid decline since ~3.3 ka. These insolation-driven EASM trends were punctuated by two millennial-scale weakening events which occurred synchronously to the cold Younger Dryas and at ~9.5-8.5 ka, and by two centennial-scale intervals of enhanced (weakened) monsoon during the Medieval Warm Period (Little Ice Age). Our precipitation reconstruction, consistent with temperature changes but quite different from the prevailing view of EASM evolution, points to strong internal feedback processes driving the EASM, and may aid our understanding of future monsoon behaviour under ongoing anthropogenic climate change.

Chronology is a necessary component of paleoclimatology. Radiocarbon dating plays a central role in determining the ages of geological samples younger than ca. 50 ka BP. However, there are many limitations for its application, including radiocarbon reservoir effects, which may cause incorrect chronology in many lakes. Here we demonstrate temporal changes in the radiocarbon reservoir age of Lake Xingyun, Southwestern China, where radiocarbon ages based on bulk organic matter have been reported in previous studies. Our new radiocarbon ages, determined from terrestrial plant macrofossils suggest that the radiocarbon reservoir age changed from 960 to 2200 years during the last 8500 cal a BP years. These changes to the reservoir effect were associated with inputs from either pre-aged organic carbon or 14C-depleted hard water in Lake Xingyun caused by hydrological change in the lake system. The radiocarbon reservoir age may in return be a good indicator for the carbon source in lake ecosystems and depositional environment.
